I would have the world domination game from that James Bond movie.
If we are talking "unlimited" budget, then I would have a racing game that has a set track, and a sit-down cabinet with a real steering wheel. I would have about 8 cabinets. They would be on a real track, and the cabinets would have wheels/brakes/motors and where you steered onscreen is where you would go on the track in real-life.
Either that, or a life-sized version of the Tron games from the movie...

Might as well throw in a bean-bag or other big soft seat, maybe a mini-fridge, too.
But I don't like to be crowded, so I'd prefer it to be a pretty big room. Basements are fun for this, I think.
I'd have a giant bouncy house and bouncy lazer tag room.
I'd also have some kind of helium + zero gravity room where we could play a game of dodgeball.
I'd love to include at least some kind of air hockey and casino room as well.
Perhaps, a mini "Price Is Right" entertainment stage would be cool, and maybe some kind of 3-d video games with lazer lights, disco balls, fog machine, strobe lights, and an old-school skating rink with disco attire featuring the 70s era.
I'd also have some kind of surround sound muzac.
Oh, and can't 4get the skeeballz, whack-a-mole, and an icee machine that serves pina coladas. That'd be awesome..
If I had unlimited space and money I'd probably just get everything.
And to be more specific:
The whole "games room" would be arranged in modules.
Like in the movie cube.
You enter one room and have to play a round of the specific game or activity in there before you can leave the room.
The whole structure would be moving and alternating positions so that every time you open a door you end up in a random location.
Games/activities would include:
Far spitting from Monkey Island
A giant chamber with huge elecromagnets and you in there in an ironsuite and a remote to control the magnets intensity.
50 different Rollercoasters
500 Slides, Waterslides, Bobslides, zomg I get really excited now!
Wooot so much stuff
EVERY fun game and activity that ever has been invented.
When I get sad I stop being sad and be awesome instead. True story.

12 top end computers divided in two seperate booths (for some 6 vs 6 teamplay fragging), in the third booth a 5 monitor multi-box system with crazy amount of bandwidth and ram (for multiboxing, obviously. >.> ). A fourth booth housing a tv with all game consoles.
A bar, a pool table and a snooker table, some dartboards and various classic arcade systems, and a sitting area with a slouchy couch around a fireplace.
A bowling alley and a huge swimming pool with currents, slides and fake jungle plants. I love fake jungle plants.
And an outside area to play some soccer or other outdoor sports.
Bar with dozens of types of whisky/beer/cognac and other types of drinks.
A concert piano.
A huge table with a dozen computers.
A bookcase with every literature masterpiece ever written.
A pool table.
A swimming pool
A cigar bar.
Huge ass television and a very comfortable couch to watch it from.
A shooting range.
A big stereo set.
All music ever produced on cd.
A dozen or more playmates coming over from Hugh Hefner's house!
A collection of guitars and basses.
Turntables and mixing equipment.
A computer specifically used for making and recording music.
